چکیده

We describe a method for calculating the absorptivity (in terms of substrate consumed) of the colored solution obtained when o-dianisidine dihydrochloride is oxidized by ceruloplasmin. By oxidizing o-dianisidine dihydrochloride with known amounts of hydrogen peroxide we could determine that the molar reacting ratio of o-dianisidine to hydrogen peroxide is 2:1. So calculated, absorptivity is 9.6 ml zmor1 cm’, the figure used to estimate ceruloplasmin oxidase activity in terms of International Units.

Automated determination of serum ceruloplasmin activity with o-dianisidine dihydrochloride as substrate.

An automated method for the enzymatic determination of ceruloplasmin with o-dianisidine dihydrochloride as substrate is described. The method enables the measurement of 30 samples per hour with a coefficient of variation (day-to-day) of 2.8%. Results correlate well (r equals 0.99 with those obtained by the corresponding manual method

Measurement of ceruloplasmin from its oxidase activity in serum by use of o-dianisidine dihydrochloride.

We have developed a new method for determination of serum ceruloplasmin from its oxidase activity, in which o-dianisidine dihydrochioride is used as substrate. o-Dianisidine dihydrochloride is more stable than is the more widely used p-phenylenediamine substrate, and forms a stable product that is measured at 540 nm. Correlation was good between results of this method and those obtained with bo...

A micromethod for the determination of ferroxidase (ceruloplasmin) in human serums.

A sensitivemethod for the detection of ceruloplasmin (ferroxidase) in human serums, using Fe(Il) as the substrate, is described. Initial Fe(ll) concentrations of 30 and 120 p.M may be used, depending on the sensitivity of the spectrophotometer, for the determination of the rate of Fe(Ill) -transferrin formation following the oxidation of Fe(lI) by ceruloplasmin (ferroxidase). Advantages of this...

Truncating mutations in the Wilson disease gene ATP7B are associated with very low serum ceruloplasmin oxidase activity and an early onset of Wilson disease

BACKGROUND Mutations in the gene ATP7B cause Wilson disease, a copper storage disorder with a high phenotypic and genetic heterogeneity. We aimed to evaluate whether 'severe' protein-truncating ATP7B mutations (SMs) are associated with low serum ceruloplasmin oxidase activities and an early age of onset when compared to missense mutations (MMs). METHODS The clinical phenotype of 59 geneticall...

Automated measurement of serum ferroxidase activity.

A method is described for automated measurement of serum ceruloplasmin ferroxidase activity. In this method, Fe2+ ions are used as the substrate. In addition, a new calibration system without ceruloplasmin is also presented. Optimum assay reaction conditions were determined. Maximal catalytic activity was obtained at 0.45 mol/L acetate buffer, pH 5.8. The reagents and calibrator are stable for ...
